Analyzing LPS in Glucose Media

The gram negative bacteria Achromobacter xylosoxidans is an opportunistic pathogen that utilized lipopolysaccharide (LPS) as an endotoxin. My previous project involved growing the 72 gene knockout mutants in different glucose concentrations to determine changes in the LPS by SDS-PAGE gel. I was responsible for two differing projects, the first prompted by contamination or labeling issue with the mutants 66-71 in the glucose trials, leading to the current project to grow these strains under new labelling (mutants 80-85, 90-95, 100-105) and analyze their structure by LC-MS. The second project was to complete time course growths of Achromobacter xylosoxidans and Alcaligenes faecalis for LPS analysis through SDS-PAGE and analysis through LC-MS to determine the growth of a specific glycan.
